Hallum, J. V. (University of Pittsburgh, Pittsburgh, Pa.), and J. S. Youngner. Urea and thermal mutants of poliovirus. J. Bacteriol. 91:2305-2308. 1966.-Thermally selected mutants of poliovirus which differed in sensitivity to heating at 50 C were inactivated with urea. With 3 m urea, urea resistance paralleled thermal resistance. In contrast, when urea-resistant mutants were selected from thermosensitive, urea-sensitive, wild-type poliovirus, the 50 C sensitivity of the selected mutants did not change. These findings indicate a lack of covariance of the urea and the 50 C-inactivation markers of poliovirus.

Polypeptide composition of urea- and heat-resistant mutants of poliovirus types 1 and 2.
Five urea-resistant and two heat-resistant mutants of poliovirus types 1 and 2 were isolated and their structural and nonstructural polypeptides compared to those of their wild-type, parental strains in an attempt to correlate mutant phenotypes with alterations in specific capsid polypeptides. Four of the seven mutants were found to contain polypeptides which differed in molecular weight from t...
متن کامل[Dissociation and reassociation of poliovirus. I. Effect of urea on the virion].
Poliovirus is dissociated by urea in the presence of mercaptoethanol or dithiothreitol into RNA and protein(s). Differences exist in the sensitivity of poliovirus type I, II or III toward these agents. The decrease in infectivity titer of the virion is in the range of 10(9). After appropriate dissociation no infectious virions are found in urea-treated poliovirus concentrates. The released vira...
متن کاملEfficient analysis of nonviable poliovirus capsid mutants.
Nonviable poliovirus capsid mutants were studied by an efficient infection-transfection system. Phenotypically, nonviable poliovirus capsid mutants appear to segregate into three classes: those that form only protomers, those that can form pentamers, and one that can form completed virions.
متن کاملPoliovirus Particles do not Form in Preinfected Cells with Reovirus
Background and Aims: Inhibition of viral growth in coinfected cells with two different viruses has been described. This phenomenon known as viral interference can occur in several virus host systems such as interference of enterovirus infection on poliovirus vaccine strains. In this study we superinfected reovirus infected HeLa cells with poliovirus to determine if poliovirus can replicate in s...
متن کاملGuanidine-resistant poliovirus mutants produce modified 37-kilodalton proteins.
Eighteen spontaneous, guanidine-resistant mutants of poliovirus were obtained by plaque selection. Isoelectric focusing demonstrated charge changes in a 37-kilodalton protein, pX, among three of the mutants. The precursor of pX, NCVP5b , also exhibited charge changes among the three mutants. pX of 12 mutants was also examined by peptide mapping with Staphylococcus aureus V8 protease. Nine of th...
